Tissue mixed root and nodules
Description Vector: pDNR-LIB (Clontech); Site_1: SfiIA; Site_2: SfiIB; Tissue: low-phosphate treated root, Glomus versiforme infected root, uninoculated root, Sinorhizobium meliloti T202 inoculated root and nodules (3-11 DAI), Sinorhizobium meliloti 102F51 inoculated root and nodules (3-21 DAI), Sinorhizobium meliloti 1021 inoculated root and nodules (28 DAI). Total RNA isolated using TRIzol reagent. No DNaseI digestion was performed. Total RNAs were pooled together. Double strand cDNA were synthesized from pooled RNA using SMART technology ( The prepared cDNA was normalized by cDNA denaturation/reassociation, treatment by duplex-specific nuclease (DSN) and amplification of normalized fraction by PCR. The normalized cDNA was then digested with SfiI, size-fractioned, directionally ligated into pDNR-LIB (Clontech) and electroprated into GC10 competent cells (Gene Choice). Clones were sequenced from the 5' end only. RNA provided by Maria Harrison, Sue Miller, Carrol Vance and Vagner Benedito.
